Seeing as the Pasuk goes on to write "ve'Lo Ari'ach ... ", what is the significance of "Vahashimosi es Mikdesheichem"?

Rashi: It means that the groups of Yisrael, who used to gather and travel to Yerushalayim, will cease.

Seforno: It refers to the destruction of the Beis-Hamikdash at the hand of Nevuzraden, commander-in-chief of Nevuchadnetzar?s army.

Megilah, 28a: It teaches us that even after a Shul 1 is destroyed, it is still called 'Kadosh'. Consequently, in the ruin of Shul the following are forbidden: Making a Hesped, manufacturing ropes, placing traps, spreading fruit on the roof to dry and using it as a short-cut.

What are the connotations of "ve'Lo Ari'ach be'Re'ach Nichocheichem"?

Seforno, Targum Onkelos and Targum Yonasan: It means that Hashem would not accept there Korbanos with goodwill (due to the sins of the people, in spite of the righteous Kohanim such as the sons of Tzadok, whose Korbanos certainly went-up as a Re?ach Nicho?ach? Seforno).

Since the Pasuk reckons eight punishments, why does the Torah write (in Pasuk 28) "Sheva al Chatoseichem"?

Rashi: Because the destruction of the Chamanim is not an independent punishment; it is the culmination of the destruction of the towers and castles.

Why is "Va'hashimosi Ani es ha'Aretz" (in the following Pasuk) not an eighth punishment?

Rashi, Rashbam and Targum Yonasan: Because it is not a punishment, but a consolation 1 - that the land will be so barren that the enemy who will live there will derive no satisfaction from it. 2
