(1) In the beginning of the Sugya, Rashi (DH Terumah u'Ma'aser) explains that one does not transgress "Lo Sachsom" when the animal is working with Terumah or Ma'aser (that is, even Ma'aser Rishon) because the word "b'Disho" of the verse (Devarim 25:4) is not referring to threshing of Terumah and Ma'aser produce, since it is not common for produce to be threshed before it is winnowed (and Terumah and Ma'aser is separated after winnowing, normally). When the Gemara concludes that only to Ma'aser Sheni -- which is "Mamon Gavohah" and belongs to Shamayim -- does the prohibition of "Lo Sachsom" not apply, but it does apply to Ma'aser Sheni, Rashi (DH Mamon Gavohah) gives a different reason for why one does not transgress "Lo Sachsom" -- it is because such produce is not fit to be eaten by the animal (and thus it cannot be called "b'Disho," as Tosfos DH v'ha'Dashos says).
